Rotary Fiber Photometry System

Recording fluorescence related to neuronal activity within the brains of freely moving tethered laboratory animals is often hindered by transmission losses and signal fluctuations resulting from rotary joint turns. To mitigate these challenges, we strategically position the Fluorescence Mini Cube with integrated LEDs and detectors at the bottom of a rotating part of a 24-channel Assisted Electrical Rotary Joint. This placement allows us to enhance overall optical transmission and effectively eliminate rotation-related artifacts.
Moreover, the rotary joint features a central hole or lumen, facilitating the passage of an optical fiber for optogenetic stimulation and/or a tube for liquid delivery.
The Rotary Fiber Photometry System is highly adaptable. It can be tailored to support alternative light sources, such as laser diodes, and accommodate various fluorophores, ensuring versatility for different experimental setups.

The Rotary Fluorescence Mini Cube for GCaMP has 4 ports, two integrated LEDs, and one integrated Fluorescence Detector. It records calcium-independent GCaMP fluorescence, excited by 405, 415, 425, or 440 nm light (isosbestic point), and its calcium-dependent fluorescence, excited by 470 nm light, with a single photodetector. The calcium-dependent GCaMP's fluorescence emission is separated from calcium-independent emission by lock-in amplifier detection or by interleaved excitations.

The Rotary Fluorescence Mini Cube with 6 ports-FRET GCaMP Isosbestic & Functional Excitations, & Red Fluorophore Fluorescence contains three integrated LED and two integrated Fluorescence Detectors. This cube can be used to detect the fluorescence from two calcium indicators and the GCaMP isosbestic point. The separation of functional and isosbestic fluorescnece signals of GCaMP is possible if the light source are modulated using lock-in demodulation or sequential detection.
